Toxicological and teratological studies with 1395-TH were performed. The observations are summarized as follows. 1. LD_50 of 1395-TH given by various routes to mice, rats and rabbits is shown in Table-1. 2. In the subacute and chronic toxicity test with 1395-TH repeatedly administered to rats by a stomach tube, the medication was done in 100, 200, 300 and 350mg/kg daily for 3 and 6 months. In the group of 350mg/kg, 5 rats out of 20 died by the 5th day, and the death may possibly have been due to low blood sugar caused by 1395-TH. The weight gain was restrained in groups more than 200mg/kg after about 3 months. No remarkable change, but low blood sugar was found in hematology and chemical and biochemical analyses of the blood. The histological examination proved no influence of 1395-TH except atrophy of the white pulp of the spleen and fatty accumulation in the adrenal cortex. 3. Teratological studies on mice and rat: The resorption and retardation of growth and ossification were observed in fetuses of mice and rats, and malformations (cleft palates and exencephalies) as well as vertebral defects which could be attributed to low blood sugar only in fetuses of mice.
