Application of the superimposing method was planned for the collection of light microscopic (LM) and scanning electron microscopic (SEM) images which were taken by the observing method of the same and fixed optical field of LM and SEM. Arrangement and establishment of the technical procedures were attempted by using some biological materials. For examples, a good result could be obtained in the microscopy of chromosome. Namely, a typical band image of the chromosome of FL cell by LM was also observed as the cubical fine structure by SEM. Application of superimposing method facilitated the collation of both LM and SEM images of the chromosome and increased its interpretable degree. Therefore superimposing method can be used as an excellent technique in the imagetreatment method for colletion of both images in LM and SEM studies.
