Effects of Tumor Necrosis Factor-α on Adherence and Motility of Squamous Cell Carcinoma Cells

Effects of recombinant human tumor necrosis-α (rTNF-α) on the adhesiveness and motility of SKG-IIIa cells, a cell line derived from cervical cancer were studied. TNF-α significantly enhanced release of fibronectin from SKG-IIIa cells. Immunohistochemical studies using an antibody against fibronectin demonstrated that the intensity of staining was increased in the TNF-α treated cells. The adherent capacity to plastic plate was significantly increased after TNF-α treatment. The migration capacity was decreased after TNF-α, which was completely blocked by simultaneous addition of an antibody against fibronectin. These results indicated thatTNF-α influenced biological behaviors of SKG-IIIa cells (e.g. adherent capacity and migration potential) , which would be mediated through the increase of fibronectin products on the cellular membrane.
