    The present study examined the utility of lymphokine activated killer(LAK) cells derived from mononuclear cells(MNCs) of lymph nodes stimulated with subcutaneously injected OK-432. LAK cells were induced by 100JRU/ml of rIL-2 from the MNCs of the popliteal and axillary lymph nodes of mice stimulated with OK-432(OK-432 group) or saline alone(control group). The number of MNCs in the OK-432 group(17.7×10^6) significantly greater than that in the control group(3.9x10^6). The anti-YAC-1 activity and anti-EL-4 activity of LAK cells induced by 3 days of incubation in the OK-432 group were significantly higher than those in the control group. Phenotypic analysis demonstrated that the percentage of asialo GM1 positive lymphocytes of LAK cells in the OK-432 group was significantly higher than that in the control group, whereas the percentage of Thy1.2, L3/T4 and Ly2 positive cells in the OK-432 group was significantly lower than that in the control group. Both anti-YAC-1 and anti-EL-4 activities were strongly inhibited by treatment with anti-asialo GM1 serum plus complement. Thus, the effector cells of LAK cells were characterized as Thy1.2 and asialo GM1 positive cells. Winn's neutralization assay against Meth A tumor cells showed that the anti-tumor effect of LAK cells in the OK-432 group was not much greater than that in the control group. These results suggest that adoptive immunotherapy using LAK cells derived from MNCs of lymph nodes stimulated with subcutaneously injected OK-432 may be useful for cancer therapy.
