The protective effect of fosfomycin (FOM) against ciclosporin (CYA)-induced nephrotoxicity was studied. Male Wister rats weighinf 250g were divided into 4 groups. Group 1 was given olive oil orally and saline inrtapertioneally. The other groups were treated by oral administration of 50mg/ke/day of CYA for 7 days. Group 2, 3 and 4 were given intraperitoneal injection of salline, 125mg/kg/day of FOM and 250mg/kg/day of FOM respectively. After these treatments, the rats were placed in metabolic cages for 24 hours, and urine and blood were collected. Then the rats were sacified for loght microscopic examination of bilateral kidneys. Blood urea nitrogen (BUN), serum creatinine(s-Cr) and urinary N-acetyl-β-glucosaminidase (NAG) activity were measured. The 24-hours creatinine clearance(Ccr) was also determined. In group 2, the level of BUN, s-Cr, Ccr and NAG activity were significantly impaired conpared to group 1. No significant diffeences were, however, found among CYA-administered groups. Light microscopy revealed the deposition of PAS-positive granules in, exfoliation of, disapperanve of brush border from and cacuolization of proximal tubular epithlium. These changes were observed likewise in all CYA-treated groups. These results revealed CYA-induced nephrotoxicity but did nor suggest the protective effect of FOM…
