血管平滑筋のCa^{2+}依存性収縮とは対照的に,スフィンゴシルホスホリルコリン(SPC)/ Rhoキナーゼ(ROK)経路を介したCa^{2+}非依存性血管収縮は,血管攣縮などの異常な血管過収縮の一因である.血管攣縮を予防するため食品由来成分を中心に探索した結果,柑橘由来成分であるノビレチンを見出した.ノビレチンはSPCによって引き起こされた血管異常収縮を強力に抑制し,40 mM K^{+}溶液によって引き起こされたCa^{2+}依存性血管収縮に対しては,微弱な抑制効果を示した.さらに,細胞質Ca^{2+}濃度と張力の同時測定では,ノビレチンは細胞質Ca^{2+}濃度に影響を与えることなくSPCによって引き起こされた血管異常収縮を抑制し,40 mM K^{+}溶液によって引き起こされたCa^{2+}依存性血管収縮に対してわずかな抑制効果を及ぼした.これまでに我々は,SPC刺激によるROKの活性化,活性化したROK細胞質から細胞膜への移動,ミオシンホスファターゼ標的サブユニット1(MYPT1)とミオシン軽鎖(MLC)のリン酸化レベルの上昇を明らかにし,ROKの活性化が血管攣縮に重要な役割を果たすことを報告してきたが,本研究において,ノビレチンは,SPC刺激によってもたらされるリン酸化MYPT1およびリン酸化MLCの発現レベルの上昇を有意に著しく抑制することが明らかになった.以上の結果は,ノビレチンが血管攣縮につながる異常な血管収縮に対する新規治療薬の標的候補として有望であることを示唆している.
In contrast to Ca^{2+}-dependent contraction of vascular smooth muscle(VSM), sphingosylphosphorylcholine(SPC)/Rho-kinase(ROK)pathway, which induces Ca^{2+}-independent VSM contraction, contributed to abnormal VSM hypercontraction such as vasospasm. After extensive screening of various food-derived components, we found that nobiletin, a citrus flavonoid, which suppressed the SPC-induced Ca^{2+}-sensitization of VSM contraction with little inhibitory effect on depolarization-induced Ca^{2+}-dependent contraction by 40 mM K^{+}. Furthermore, in the simultaneous measurement of [Ca^{2+}]_{i} and force, nobiletin inhibited the SPC-induced Ca^{2+}-sensitization of VSM contraction without change of [Ca^{2+}]_{i}, but had slight effects on 40 mM K^{+}-induced Ca^{2+}-dependent contraction and [Ca^{2+}]_{i}. We previously reported that SPC induced ROK translocation from cytoplasm to cell membrane, and activation of ROK led to the increase in phosphorylation levels of myosin phosphatase target subunit1(MYPT1) and myosin light chain(MLC), suggesting that activation of ROK plays an important role in vasospasm. Nobiletin significantly inhibited the SPC-induced phosphorylation levels of MYPT1 and MLC. These results suggest that nobiletin is a promising candidate for novel therapeutic drug target for abnormal VSM contraction leading to vasospasm.
