The protective effect of prostaglandin Ej against liver tissue injury was examined in acute bleeding(1% of body weight)models of 24 hour obstructive jaundiced rats. To assess the liver metabolic changes after blood letting, we used noninvasive in vivo <31>^P-MRS. The changes in inorganic phosphate(Pi) and β-ATP peaks (expressed as the height of signals) were monitored and analyzed. We also observed the changes in the chemical shifts of Pi signals relative to α-ATP signals. Simultaneously, to assess the hepatic hemodynamics. the hepatic tissue blood hemoglobin concentration(IHb) , oxygen saturation of hemoglobin(ISO_2) , and consumption of oxygen(IC-O_2) in situ were estimated by reflectance spectrophotometry. The ratio of β-ATP/Pi in the PGE_1 treated group was significantly higher than that in the non-treated group and chemical shifts in the PGE_1 group remained relatively low after blood letting. The ISO_2, IC-O_2 values in the PGE_1 group were significantly higher than those in the non-treated group at 60 min after blood letting. These values were similar to the original values.The results of experimental studies suggest that a small amount of bleeding in obstructive jaundice is dangerous and PGE_1 may protect hepatic metabolic disorder and prevent acidotic changes of liver cells from bleeding during surgery in patients with obstructive jaundice. The effects of PGE_1 were considered to be associated with the maintenance of ISO_2, IHb and IC-O_2.
