Cultivation of Borrelia duttonii in tissue culture

Borrelia Duttonii 406K strain which has been maintained in our laboratory in mice did not grow in Kelly medium, although it has been reported that B. duttonii grows well in the dedium. Therefore, the author attempted to obtain survival or replication (or both) of 406K strain in vitro. In the present studies, 406K was shown to replicate in conventional monolayer cultures of cottontail rabbot epithelium (Sf1Ep) in Eagle minimum essential medium supplemented with 10% fetal calf serum under a microaerophilic condition (O_2 : 3%, CO_2 : 5%, N_2 : 92%) at 34℃ and has been maintained in continuous cultivation for 6 months by serial transfer of 20subcultures. The opimum concentrations of oxygen and carbon dioxide for replocation of B. duttonii at 34℃ ranged from 3 to 5%, and 5 to 10%, respectively. On the seventh day after inoculation of 10^3 to 10^5 organisms/ml, the yield of borreliae was in the range between 2.0×10^6/ml and 3.3×10^6/ml.
