Influence of 5-Fluorouracil on Respiratory Metabolism and Cell Division of Ehrlich Ascites Cancer Cell

The author investigated the anticancer effects of 5-FU on respiratory metabolism, glycolysis and cell division of Ehrlich ascites cancer (EAC), and obtained following results. 1) An action of 5-FU on EAC cells revealed to be decreasing QO_2 and Crabtree effect of EAC cells in vitro. 2) Decrement of pH in cell culture medium containing glucose was recognized, while, on the contrary, lactic acid quantity in reaction medium was increased. 3) Activities of NADH-DH and SDH of EAC cells were considerably inhibited in vitro. 4) On the mice that was intraperitoneally transplanted EAC cells, was contineously injected with 5-FU intraperitoneally after 5th of EAC cell inoculation, activity of SDH was slightly checked, but their daily changes was scarcely recognized in vivo. Concerning activity of NADH-DH, however, was recognized a tendency as same as it in vitro in the early period of 5-FU injection, on the contrary, in later period observed more remarkable increase than the control group. 5) Effect of 5-FU on cell division wes remarkably observed in the later period of contineous injection, ―increased of mitotic ratio in prophase stage, on the contrary, recognized remarkable decrease of ratio of cell division in anaphase or telophase. 6) Tumor index of treated group with 5-FU was recognized more less than control group in daily. 7) Specific action of 5-FU was clarified from above the mentioned results that effects of 5-FU on Ehrlich ascites cancer cell in vivo may be inhibit cell division until the stage of prophase, and block metabolic turnover of cell division.
