内容記述(抄録等)The effects of pancuronium (PBr) and vecuronium (VBr) on contractile response of the tracheal smooth muscle of the guinea pig to either acetycholine (ACh ; 5x10^<-9>~2x10^<-4>M/L) or carbamylcholine (CCh ; 5x10^<-10>~2x10^<-5>M/L), which are known to contract tracheal smooth muscle affecting muscarinic receptor, were investigated. In addition, the study was designed to examine the modifications of the effects of PBr and VBr by pretreatment with phentolamine (PH), propranolol (PR), diphenhydramine (DH) and indomethacin (IM). PBr (5x10^<-7>~10^<-5>M/L) shifted the dose response curve of ACh to the right in a dose related manner. With PBr 10^<-5>M/L, EC_50 values (the concentration producing 50% of maximum concentration) for ACh and CCh responses were increased. The muximum concentrations produced by ACh were also increased, while that produced by CCh was not affected by PBr. VBr 10^<-5>M/L shifted the dose response curve of ACh to the left, producing a decrease in EC_<50> and an increase in the maximum concentration. VBr failed to alter the contractile response to CCh. These results indicate that PBr antagonizes the muscarinic receptor medicated contraction of the tracheal smooth muscle and possesses anticholinesterase-like effect. It is also suggested tha VBr has anticholinestrase-like effect. When tracheal smooth muscke was pretreated with PH or PBr, PBr produced an increase in the maximum contraction induced by ACh. It could be possible that the effects of PBr on tracheal smooth muscle may be, at least in part, attributed to the α-or β-receptor mediated response. When the tracheal smooth muscle was pretreated with DH or IM, PBr produced a decrease in EC_<50> and in the maximum contraction induced by ACh, respectively. This may suggest that the inhibitory effect of PBr on the muscarinic receptor was curtailed by the pretreatment with DH and that anticholinesterase-like effect of PBr was inhibited by the pretreatment with IM. Since VBr changed neither EC_<50> nor the maximum contraction induced by ACh, it is unlikely that the effects of VBr on tracheal smooth muscle are related to the responses mediated by the endogenously released substances (α-, β-adrenergic agonists, histamine or prostanoid). In conclusion, PBr and VBr affect the contractile reponses of the airway smooth muscle to cholinergic agonists, Ach and CCh. Thus, when large amount of these muscle relaxants are administered, precaution against the airway constriction is necessary in the patients with bronchial asthma and/or airway hypersensitivity.
