内容記述(抄録等)Mice undergoing withdrawal after chronic treatment with ethanol exhibited lowered seizure thresholds to β-DMCM, a benzodiazepine inverse agonist and to NMDA, a glutamate receptor agonist, but not to bicuculline, a GABA_A receptor antagonist; picrotoxin, a Cl? channel blocker; or to caffeine, an adenosine receptor antagonist. These results suggest that the GABA_A-Cl? channel complex and central purinergic systems may not be involved in the lowering of convulsive thresholds induced during withdrawal from chronic ethanol administration. The lowering of seizure threshold of β-DMCM occurred 48-72 h, and that of NMDA 24-72 h, after the last administration of ethanol. Β-DMCM-induced convulsions in control group were inhibited by CPP and MK-801, which are competitive and noncompetitive NMDA antagonists, respectively. The inhibitory effect of CPP was antagonized by a low dose of NMDA, which by itself did not induce convulsions. On the other hand, convulsions produced by NMDA in control animals were not affected by low doses of β-DMCM or diazepam, both benzodiazepine agonists, or flumazenil, a benzodiazepine antagonist. However, in the ethanol treated group, the NMDA convulsions were potentiated by β-DMCM and inhibited by diazepam. The stimulatory effect of β-DMCM and the inhibitory effect of diazepam were antagonized by flumazenil. These results suggest that ethanol withdrawal potentiates the convulsive effects of β-DMCM and NMDA. The stimulatory effects during withdrawal from chronic ethanol treatment may be, in part associated with an activation of the NMDA Ca++hannel complex via the β-carboline recognition site. In control animals the convulsive effects of the β-carboline may be mediated by activation of the NMDA system, but that produced by NMDA may not involve the β-carboline recognition site.
