内容記述(抄録等)Incidence of amyloid deposits was surveyed in 3,000 cows at Ube Abattoir during a four-year period from 1981 to 1984. At autopsy the kidneys were examined by the method of iodine sulfuric acid reaction as a screening test for amyloid deposits. The dark-brown areas (positive reaction) were found in 23 out of 3,000 coes ; 22 out of 1,800 Holstein cows and one out of 1,200 Japanese black cows. Amyloid deposits in various organs from these 23 cows were studied histologically, immunohistochemically, and ultrastucturally. Histologically amyloid deposits were detected in all of 23 cows (100%) in the kidney, 13 out 22 (59.1%) in the liver, 12 out of 22 (54.5%) in the adrenal gland, 10 out of 22 (45.5%) in the spleen, 14 out of 22 (63.6%) in the intestinal tract, and 2 out of 22 (9.1%) in the heart. The kidney, liver and adrenal gland mecroscopically altered in color of each organ in which amyloid depesits were demonstrated more than 20% of the whole tissue dimensions using point count technique. Immunohistochemically amyloid deposits reacted with an anti-human amyloid A antibody. On electrom microscopy, amyloid was composed of non-branched, straight fine fibrils measuring 12nm in width. These results indicate that the incidence of amyloidosis in Holstein cows is higher than in Japanese black cows, and confirm that the amyloid protein in coes is amyloid A protein similar to that in secondary amyloidosis.
