内容記述(抄録等)In recent years, several reports on the observation of the luminal surface of aorta using scanning electron microscope have been reported, but any of them failed to present the fine structure of the endothelial cell. The authors made some improvements upon the routine methods of preparation of specimens for electron microscopic research, and could observe the fine structure of the endothelial surface of aorta in dogs. The findings obtained were as follows : 1) By the single usage of any one of the fixatives such as formaldehyde, ethanol, acetone, Zenker's solution, Carnoy's solution, glutaraldehyde, and osmic acid, the authors could never have a clear finding of the structure of the luminal surface ; but using either the doudle fixation method by glutaraldehyde and Dalton's solution or the SUMP (Suzuki's Universal Micro-Printing) method, the authors could succeed to find clearly its fine structure. And the dehydration using isoamyl acetate after tissue fixation could prevent the deformation of the luminal surface structure. 2) The scanning electron microscopic findings hitherto reported were not of the luminal surface but the subendothelial tissue. 3) the endothelial cell was elongated and distorted in polygonal shape, and measured 38~56μ×10~14μ. An oval shaped nucleus (measuring 9~12μ×7.5~8.4μ) existed in the center of the cell. The adjacent cells were brought together into a tight contact one another, building up the smooth luminal surface of the vessel. The endothelial cell had a tendency to arrange themselves along the longitudial axis of the vessel. There could be found either intercellular bridge-like structures nor linear endothelial folds.
